Abstract
Mitophagy is a selective form of autophagy for removal or clearance of damaged mitochondria via the autophagy-lysosome pathway. Among various mitophagy regulatory mechanisms, PINK1, a protein kinase, and Parkin, an E3 ligase, are two critical players in control of mitophagy, with important implications in neurodegenerative disorders such as Parkinson's disease (PD). In our recent studies, we aimed to investigate the upstream regulatory mechanisms of PINK1. I will present some of our unpublished data, from the transcriptional regulation of PINK1, to a novel role of G6PD in regulation of PINK1 stabilization and kinase activation in response to mitochondrial damage. In addition, I will discuss a novel function of a well-established autophagy inhibitor, Spautin-1 in regulation of mitophagy, via PINK1. Our results provide a deeper insight into the molecular mechanisms in control of PINK1, the guardian of mitochondria and lay foundation for development of novel interventional strategies in PINK1 and mitophagy-related human diseases such as neurodegeneration and cancer.
Biography:
Dr. Han-Ming Shen is currently a Chair Professor and Associate Dean (Teaching), Faculty of Health Sciences, University of Macau, and Visiting Professor, Yong Loo Lin School of Medicine, National University of Singapore. He received his Bachelor of Medicine, Master of Medicine and PhD from Zhejiang Medical University and National University of Singapore, respectively. His research is focused on the autophagy-lysosome pathway, mitophagy, as well as on glucose metabolism in cancer cell biology. Dr. Shen has published more than 250 papers, with more than 44,000 citations and H-index at 103 (Google scholar). Currently, he serves as the Associate Editor for Autophagy, and as member of editorial board for several other journals such as Burns & Trauma, Life Metabolism, and Aging Cell.
